LEBS TECHNICAL FACILITY
The study of the fundamental mechanisms of protein-protein and protein-nucleic acid interactions requires high-tech equipment of biochemistry and biophysics of proteins. LEBS has a technical facility combining several methodologies for the study of physico-chemical properties of biological macromolecules and their complexes. 3 platforms and are functional:
• Cloning and Mutagenesis Platform. Since December 2007, we provide services in molecular biology. The acquisition of a pipetting robot epMotion has allowed automation of cloning steps, expression tests and purification protocols by combining parallelization, miniaturization and standardization. In addition, LEBS has many other biochemistry equipment needed to obtain pure protein (incubators, breaker cell streaming, FPLC-Aktas, microvolume spectrophotometer,...).
• Crystallization Platform. Established in 2006, the platform has two crystallization robots, one visualization robot, and several microscopes coupled to computers, that facilitate structural studies of proteins via the automation of crystal growth. Users can use the robots by themselves after training or they can take advantage of the platform staff expertise, in service mode or in collaboration from the crystallisation to the crystal structure resolution. The visualization robot is freely accessible and scans are visible and programmable remotely. The proximity of the Soleil synchrotron is also a considerable asset for protein crystallography at LEBS.
• Biophysics Platform. LEBS has several essential equipment for biophysical characterization of proteins and protein complexes. Some of them are: circular dichroism, SEC-MALS, analytical ultracentrifugation, microcalorimetry, fluorometer, fluorescence microplate reader, polarization anisotropy, Biacore, ... the first 4 ones are open to external users.
